Design and engineering of tumor-targeted, dual-acting cytotoxic nanoparticles

چکیده

The possibility to conjugate tumor-targeted cytotoxic nanoparticles and conventional antitumoral drugs in single pharmacological entities would open a wide spectrum of opportunities nanomedical oncology. This principle has been explored here by using CXCR4-targeted self-assembling protein based on two potent microbial toxins, the exotoxin A from Pseudomonas aeruginosa diphtheria toxin Corynebacterium diphtheriae, which oligo-floxuridine monomethyl auristatin E respectively have chemically coupled. resulting multifunctional hybrid nanoconjugates, with hydrodynamic size around 50 nm, are stable internalize target cells biological impact. Although chemical conjugation minimizes activity partner complexes, concept drug combination proposed is fully feasible highly promising when considering multiple treatments aimed higher effectiveness or facing therapy cancers acquired resistance classical drugs.
